A home-built top-loading high-pressure cell (pressure range 0.1 to 350 MPa) is associated with Koala fluorimeter from ISS, Inc (Champaign, IL) capable of single point intensity measurements vs pressure or vs time after pressure change, as well as pressure-dependent automated excitation and emission spectra of all UV and Vis excitable dyes.
The system is run using an automated HUB 440 pressure generating system from Pressure BioSciences (South Easton, MA). High Pressure Sample holder is temperature regulated using a water bath (2-80 deg C) and optics are cleared with nitrogen gas. Inner sample holder is 500 uL quartz cell closed with Dura-Seal and an O-ring. Pressure transducing liquid is 18 MOhm purified water.
